α-propargylhomoterephthalic acid dimethyl ester is a cytotoxic compound that has been shown to have antitumor activity. It is a prodrug of pralatrexate and can be hydrolyzed to active form by esterases or glucuronidases. The active form exhibits cytotoxicity against tumors and is effective in killing cells with low glutathione content. 

Pralatrexate is an antifolate for the treatment of relapsed or refractory peripheral T-cell lymphoma. It is also being studied for other types of lymphoma and solid malignancy such as non-small-cell lung cancer, breast cancer, and bladder cancer.
